Salinity influences disease-induced mortality of the oyster Crassostrea gigas and infectivity of the ostreid herpesvirus 1 (OsHV-1) ArchiMer
Fuhrmann, Marine; Petton, Bruno; Quillien, Virgile; Faury, Nicole; Morga, Benjamin; Pernet, Fabrice.
Mortality of young Pacific oysters Crassostrea gigas associated with the ostreid herpesvirus 1 (OsHV-1) is occurring worldwide. Here, we examined for the first time the effect of salinity on OsHV-1 transmission and disease-related mortality of C. gigas, as well as salinity-related effects on the pathogen itself. To obtain donors for OsHV-1 transmission, we transferred laboratory-raised oysters to an estuary during a disease outbreak and then back to the laboratory. Oysters that tested OsHV-1 positive were placed in seawater tanks (35‰, 21°C). Water from these tanks was used to infect naïve oysters in 2 experimental setups: (1) oysters acclimated or non-acclimated to a salinity of 10, 15, 25 and 35‰ and (2) oysters acclimated to a salinity of 25‰; the...

Molecular responses of Ostrea edulis haemocytes to an in vitro infection with Bonamia ostreae ArchiMer
Morga, Benjamin; Arzul, Isabelle; Faury, Nicole; Segarra, Amelie; Chollet, Bruno; Renault, Tristan.
Bonamiosis due to the parasite Bonamia ostreae is a disease affecting the flat oyster Ostrea edulis. B. ostreae is a protozoan, affiliated to the order of haplosporidia and to the cercozoan phylum. This parasite is mainly intracellular, infecting haemocytes, cells notably involved in oyster defence mechanisms. Suppression subtractive hybridisation (SSH) was carried out in order to identify oyster genes differentially expressed during an infection of haemocytes with B. ostreae. Forward and reverse banks allowed obtaining 1104 and 1344 clones respectively, among which 391 and 480 clones showed a differential expression between both tested conditions (haemocytes alone versus haemocytes in contact with parasites). ESTs of interest including genes involved in...

A production monitoring network focused on the blue mussel Mytilus edulis, was developed at the Charentais sounds' geographic level to assess the spatial and temporal variability of the mussel production. Similarly to the oyster growth monitoring network REMORA, an homogeneous and calibrated mussel population (L = 35.5 mm, TW = 3.35 g) was distributed into six rearing sites, representative of the sounds production. The rearing technics were compared and intercalibrated with previous growout facilities used for preliminary trials (round bags-oyster bags). The monitoring was based upon biometric and biochemical measurements on a 3 month and monthly basis, for ail and 3 sites respectively. The mortality rates showed a significant spatial and temporal...

Background: Massive mortality outbreaks affecting Pacific oyster (Crassostrea gigas) spat in various countries have been associated with the detection of a herpesvirus called ostreid herpesvirus type 1 (OsHV-1). However, few studies have been performed to understand and follow viral gene expression, as it has been done in vertebrate herpesviruses. In this work, experimental infection trials of C. gigas spat with OsHV-1 were conducted in order to test the susceptibility of several bi-parental oyster families to this virus and to analyze host-pathogen interactions using in vivo transcriptomic approaches. Results: The divergent response of these oyster families in terms of mortality confirmed that susceptibility to OsHV-1 infection has a significant...

Suppression substractive hybridisation (SSH) and real time PCR reveal differential gene expression in the Pacific cupped oyster, Crassostrea gigas, challenged with Ostreid herpesvirus 1 ArchiMer
Renault, Tristan; Faury, Nicole; Barbosa Solomieu, Valérie; Moreau, Kevin.
Virus-induced genes were identified using suppression subtractive hybridisation (SSH) from Pacific cupped oyster, Crassostrea gigas, haemocytes challenged by OsHV-1. A total of 304 clones from SSH forward library were sequenced. Among these sequences, some homologues corresponded to (i) immune related genes (macrophage express protein, IK cytokine, interferon-induced protein 44 or multicopper oxidase), (ii) apoptosis related genes (Bcl-2) and (iii) cell signalling and virus receptor genes (glypican). Molecular characterization and phylogenic analysis of 3 immune-related genes (macrophage expressed protein, multicopper oxidase and immunoglobulin domain cell adhesion molecule) were performed. Finally, quantitative PCR revealed significant changes in the...

Can survival of European flat oysters following experimental infection with Bonamia ostreae be predicted using QTLs? ArchiMer
Harrang, Estelle; Heurtebise, Serge; Faury, Nicole; Robert, Maeva; Arzul, Isabelle; Lapegue, Sylvie.
The present study identifies quantitative trait loci (QTLs) in response to an experimental infection with the parasite responsible for bonamiosis, Bonamia ostreae, in two segregating families of the European flat oyster, Ostrea edulis. We first constructed a genetic-linkage map for each studied family and improved the existing genetic-linkage map for the European flat oyster with a set of SNP markers. This latter map now combines the best accuracy and the best estimate of the genome coverage available for an oyster species. Secondly, by comparing the QTLs detected in this study with those previously published for O. edulis in similar experimental conditions, we identified several potential QTLs that were identical between the different families, and also...

Faury, Nicole; Ratiskol, Jacqueline.

Immunohistochemistry (IHC) assays were conducted on paraffin sections from experimentally infected spat and unchallenged spat produced in hatchery to determine the tissue distribution of three viral proteins within the Pacific oyster, Crassostrea gigas. Polyclonal antibodies were produced from recombinant proteins corresponding to two putative membrane proteins and one putative apoptosis inhibitor encoded by ORF 25, 72, and 87, respectively. Results were then compared to those obtained by in situ hybridization performed on the same individuals, and showed a substantial agreement according to Landis and Koch numeric scale. Positive signals were mainly observed in connective tissue of gills, mantle, adductor muscle, heart, digestive gland, labial palps, and...

Detection and distribution of ostreid herpesvirus 1 in experimentally infected Pacific oyster spat ArchiMer
Segarra, Amelie; Baillon, Laury; Faury, Nicole; Tourbiez, Delphine; Renault, Tristan.
High mortality rates are reported in spat and larvae of Pacific oyster Crassostrea gigas and associated with ostreid herpesvirus 1 (OsHV-1) detection in France. Although the viral infection has been experimentally reproduced in oyster larvae and spat, little knowledge is currently available concerning the viral entry and its distribution in organs and tissues. This study compares OsHV-1 DNA and RNA detection and localization in experimentally infected oysters using two virus doses: a low dose that did not induce any mortality and a high dose inducing high mortality. Real time PCR demonstrated significant differences in terms of viral DNA amounts between the two virus doses. RNA transcripts were detected in oysters receiving the highest dose of viral...

A study of autophagy in hemocytes of the Pacific oyster, Crassostrea gigas ArchiMer
Picot, Sandy; Morga, Benjamin; Faury, Nicole; Chollet, Bruno; Dégremont, Lionel; Travers, Marie-agnes; Renault, Tristan; Arzul, Isabelle.
Macroautophagy is a mechanism that is involved in various cellular processes, including cellular homeostasis and innate immunity. This pathway has been described in organisms ranging in complexity from yeasts to mammals, and recent results indicate that it occurs in the mantle of the Pacific oyster, Crassostrea gigas. However, the autophagy pathway has never been explored in the hemocytes of C. gigas, which are the main effectors of its immune system and thus play a key role in the defence of the Pacific oyster against pathogens. To investigate autophagy in oyster hemocytes, tools currently used to monitor this mechanism in mammals, including flow cytometry, fluorescent microscopy and transmission electron microscopy, were adapted and applied to the...

Field characterization of summer mortality was performed in France in the frame of the Morest project. Natural and hatchery spat were compared between three oyster production areas in France. Regardless of the natural or hatchery origin, oysters died during the reproduction period after temperature reaches 19 degree C. Thus, in southern areas, temperature accelerated gametogenesis of small spat (10mm) as well as adults, and mortality appeared for the two age classes. In contrast, sexual maturation proceeded more slowly in northern where spat mortality was lower compared to 18 months old oysters. However, critical gametogenesis and temperature were not sufficient to induce mortalities, as observed in examples with stable environment. Alternatively sediment...

Soletchnik, Patrick; Le Moine, Olivier; Goulletquer, Philippe; Geairon, Philippe; Razet, Daniel; Faury, Nicole; Fouche, Dominique; Robert, Stephane.
